Day 16 thoughts. Phagefinders.
Planned: Need to get samples to EM facility, early. Notebook check today; examine paragraphs written by students. Chiara, Lazlo. No phage? WJ phage given.
Amanda: Needs to pick from the newly found plate that has been parafilmed.
Did:
order of business:
1) which samples are ready for EM analysis? HT phage to EM facility.
2) room cleanliness.
3) written paragraphs are due.
4) Last week and one-half, we will be doing DNA isolation together. Next two weeks, phage purification.
5) Kristen not here at end of week--what will be left on hold?
6) Students look at their plates, all samples, while I am taking samples to EM. Be able to tell me what needs to be done by the time I return.
7) Labeling and use of soil samples collected at Central Park, from last Friday.
8)
Me pre students: email Raj, Zara.
Kristen has HT 1st generation A, HT B2, CH1 first generation, 2nd generation C. The A preparation also has C. CH1 is looking pure,
Chris has made HT of his phage Sample 1 at F2...He also made HT sample 19 today, at F1. These have not gone to EM, however F1 of sample 1 did go to EM. F2 plates for sample 19, one with phage already on it, so HT of F2 sample 19 can be made tomorrow.
Steve has HT from 8, he is concerned that it is 2 phages, EM.
since plates were left out at RT over the weekend, it is recommended that they be photographed and compared to previous plaque morphology. We believe that the phage may have a different phenotype at the alternate temperature.
