Planned:  1-3:00 pm today; students will be visiting EM facility re: HT phage. 

Address incubator function.  Old plates, tips, racks to dishwashing. 

Genevieve: needs to pick plaque from 10-4 of her garden phage, and this plating will be F2 once removed.  She is also propagating 2 new samples

Kristen: yesterday, diluted 2 plaques out.  Shooting from her 2nd generation from these, and she won't be here Wed. so stuff needs

Amanda:  picked 2 separate phages with similar plaque morphologies, will pick Chris extra phage tomorrow. 

Lazlo: will just be waiting because his single phage that he has picked 2 separate plaques, his air-shaft sample was plated yesterday.  PT 1 and 2 had only 1 day growth before 4 C, he may have plaques there.

Lazlo:  PT 1 and 2 did not have plaques yet, he is giving them another day and therefore has a day to lay low. 

Therefore, Lazlo will be working with the program clone, today.  He will be importing the D29 sequence into the program and digesting the DNA with BSe II and Cla I, predicting the band sizes.

Chris:  Plating sample 18 (going for a 3rd phage, from his house, this sample is documented in the whole table.)  He will also be in the EM facility today. 

Kristen:  Going to EM today, leaving early to catch flight, missing first hour of 14th.  Making chart of all samples, generations, making list of things will be doing on Monday.  B1, B2 in incubator and CH1.  We are to take these out, parafilm them and place in fridge.  She is trying to make lacy plates with all 3, all are F2, therefore they can be harvested when she comes back.  Clear Plack in Ch1 is only F1, it will be picked on Monday so that it can be diluted and propagated further.  A, in the F2 generation is with C and isn't separated.  A is therefore only at HT F1.  A has been plated at F2 and still has C, so it has been re-picked and is growing right now and will be plated again today, it will ...Kristen is worried that her C that was preppred for EM was unfiltered, perhaps we will see smeg, too!

Genevieve: picked plaque from 10-4 of her garden phage, this is now F2.  This will be Top-Agar plated today.  She is also propagating 2 new samples that will be plated today

Chiara:  Is plating her phage at F2, and will be growing them at 2 different temperatures. 

By the way, sunlight seems to cause M. smegmatis to develop pigmentation.

Electron Microscopy Visit:

We visited the EM facility for about 2 hours, while our samples were being examined and photographed.  This allowed us to visualize the imaging of several of our phages, all of which are icosahedral; however the phage tails are of various lengths and it will be important to study phage tail length with respect to plaque size formation, geographic location, soil type, etc.

The electron microscope was described, and students visited the transmission electron microscope where the samples were being imaged.  Students also visited the scanning electron microscope that was in the adjoing room, where the students benefited from Frank Macaluso's (the director of the analytical imaging facility) scientific insight.
