Day 20 thoughts. Phagefinders.
Planned:
Rhodococcus started for incubation over the weekend with 5 samples.
What would it take to annotate the genome; how is it done?
DNA reagent preparation.
Preparation of materials for autoclaving, including tips, micro-centrifuge tubes.
Did:
checked up on students questions for the next week. These were tailor made for each student; Chris's question is: Describe the location from where the phages were gathered, the nature of their plaque morphologies, what Chris saw under the EM, and if he believes they will have similar genomic sequences (reference the article)?
amanda's question was essentially the same as last weeks, since she now has phage.
Steven's job was to type up his notes, explaining his phage phylogeny.
Lazlo's woodchip sample from central park has cleared the plate, he also needs to harvest BF0316Af1 which is expected to be low high titer. Therefore, he has gone back to his eppendorf from that plaque which has been in the fridge. He will plate 0,-1,-2. Regarding his new phage CP20337, this will be replated from the soil sample at 0,-1,-2,-3.
Amanda has a big clear blotch on one of her plates, but otherwise her plates that she repicked from yesterday that were F1 (delta TNJ, PSNC) which have F1 phage didn't generate F2. She will be leaving these plates at RT over the weekend, and I recommend ...she will plate the giant plaque that she picked at -2, -4. Also, she will pick a plaque again from both of her F1's (big plaques), and plate 0,-1,-2,-3. Her giant plaque is from the pigeon bridge, hopefully this is phage she can work with even though at the F1 we only saw
Genevieve: Will leave her plates at 37 C, and Monday will be able to harvest garden phage HT at F2. It is important to note that she has an F1 high titer. She has a sample in the shaker room today that needs to be plated.
Chiara: 1) Big phage will be plated today. 2) Make HT phage. 3) photograph new phage. 4) pick new phage --> F2 (w100) It is a tough decision to see what Chiara should do
Monday, Chiara will be harvesting F2 from w100. Today, she should do same dilutions here.
