Planned:  How do plaque forming units (PFU),  as determined by spot dilution,  relate to the amount of DNA that we isolate?  We will also top-agar plate rhodococcus today (sideline experiment, hunting for phage of another bacteria), at 2 different concentrations.  This will require filtering of dirt incubated with rhodococcus for 48 h, performing the isolating phage procedure, using rhodococcus instead of smegmatis. 

AM: make rhodococcus top-agar, using LB with Noble Agar added. 

Where are my F2 phage samples for EM? 

Let's look at the DNA results from yesterday, while we do another gel analysis today.

Digestion:    Cla 1 restriction digestion.  Chris, Blake:  ran gel, 0.8% agarose. 

Paragraphs:  Bste II restriction enzyme analysis, emailed to me.  including a conversation with Lazlo. 

                        Have we found unique phage fingerprints?

                        Did we find unique phage?

                        Describe the actual Molecular Weight of your fragments, after BSTE II restriction.

Prep of phage for EM:  AECOM analytical imaging does the grid preparations for us, we give them aliquots of our phage.
